Research Interests:

The long-term goal of our research program is to understand the mechanisms by which extracellular proteases elicit cellular responses and contribute to vascular diseases and cancer progression. The focus of our work is on protease-activated receptors (PARs), which are GPCRs that signal in response to extracellular proteases. We are currently examining the novel ubiquitin-dependent processes that control PAR endocytic sorting and signal regulation in endothelial and other cell model systems. To accomplish this goal, we are using comprehensive approaches to identify the ubiquitination machinery and other novel regulators of PAR signaling and trafficking using siRNA library screens. We also study the function of PAR signaling in regulation of endothelial barrier permeability and the control of breast carcinoma transendothelial cell migration. We are also interested in understanding the function of PAR cross-talk with ErbB family members in breast carcinoma invasion and metastasis.
